Can I customize a CTA in the WordPress design theme?
I want it to change color, when pointed at, like this contact button ​https://ester.co/

Are there fully-customizable themes?

I think this would be a better request in the support forums (​https://wordpress.org/support/forum/wp-advanced/) and they should be able to assist with a solution like what you linked to. Perhaps some CSS customization and/or a theme/plugin will be the best solution.
